操作步骤：
一、开盖预处理
蛋白冻干粉 4℃平衡 30 min，12000 rpm 低温离心 5 min，将粉末离心至管底。
二、蛋白复溶
加入无菌纯水配制成 0.1~0.5 mg/mL 蛋白液（浓度不低于 0.1 mg/mL）；室温静置 5~10 min，禁止剧烈涡旋、反复吹打，避免蛋白变性。
三、C-His 标签蛋白纯化
预冷 PBS 平衡 Ni-NTA 柱；样品以 0.5 mL/min 流速上样；30 mM 咪唑缓冲液洗除杂蛋白，250 mM 咪唑洗脱目标蛋白。
四、分装保存
洗脱蛋白经 0.22 μm 滤膜除菌，无菌管分装；-20~-80℃冻存，添加 0.1% BSA 或 5% 海藻糖稳定剂，避免反复冻融。
